Cannabinoids reduce tremor associated with motor disorders induced by injuries and neurodegenerative disease. Here we show that this effect is mediated cannabinoid receptors on astrocytes in the ventral horn of spinal cord, where alternating limb movements are initiated. We first demonstrate reduced a mouse model essential after intrathecal injection analog WIN55,212-2. investigate underlying mechanism using electrophysiological recordings cord slices endocannabinoids released from depolarized interneurons activate astrocytic receptors, causing an increase intracellular Ca2+, subsequent release purines inhibition excitatory neurotransmission. Finally, anti-tremor action WIN55,212-2 cords mice suppressed knocking out CB1 astrocytes. Our data suggest cannabinoids via their Astrocytes regulate synaptic transmission. Carlsen et al. report activated inhibit transmission and, thereby, decrease tremor.
